Characterization of oligosaccharides in milk of a mink, Mustela vison OAK
Urashima, Tadasu; Nakamura, Tadashi; Ikeda, Akiko; Asakuma, Sadaki; Arai, Ikichi; Saito, Tadao; Oftedal, Olav T.; 浦島, 匡; 中村, 正.
Carbohydrates were extracted from a sample of milk from a mink, Mustela vison (Family Mustelidae). Free neutral and acidic oligosaccharides were isolated from the carbohydrate fraction and their chemical structures were compared with those of white-nosed coati (Nasua narica, Procyonidae) and harbour seal (Phoca vitulina, Phocidae) that we had studied previously. The ratio of free lactose to milk oligosaccharides was similar to that in milk of the white-nosed coati; in both species, this ratio was much lower than that in the milk of most eutherians. The neutral oligosaccharides of mink milk had alpha(1-3)-linked Gal or alpha(1-2)-linked Fuc residues at their non-reducing ends, as in the neutral oligosaccharides of white-nosed coati milk. Some of the neutral...

Characterization of two novel sialyl N-acetyllactosaminyl nucleotides separated from ovine colostrum OAK
Sasaki, Masashi; Nakamura, Tadashi; Hirayama, Kentaro; Fukuda, Kenji; Saito, Tadao; Urashima, Tadasu; Asakuma, Sadaki.
The milk/colostrum of some mammalian species is known to contain sugar nucleotides including uridine diphosphate (UDP) oligosaccharides in addition to lactose and milk oligosaccharides, but the detailed structures of these UDP oligosaccharides have not so far been clarified. In this study we isolated two UDP-sialyl N-acetyllactosamines from ovine colostrum and characterized them using H-1-NMR and MALDITOFMS spectroscopies. Their structures were found to be Neu5Gc(alpha 2 - 3) Gal (beta 1 - 4)GlcNAc alpha 1-UDP and Neu5Gc(alpha 2-6) Gal(beta 1-4)GlcNAc alpha 1-UDP.

Urashima, Tadasu; Yamamoto, Tomoko; Hirayama, Kentaro; Fukuda, Kenji; Nakamura, Tadashi; Saito, Tadao; Newgrain, Keith; Merchant, Jim; Green, Brian; Messer, Michael.
Milk oligosaccharides were separated from the carbohydrate fraction of milk of the tiger quoll a species of marsupial that is closely related to the eastern quoll, Dasyurus viverrinus. They were characterized by H-1 - nuclear magnetic resonance spectroscopy and matrix - assisted laser desorption/ionization time-of-flight mass spectrometry. The following oligosaccharides were identified; Gal(beta 1-3) Gal(beta 1-4) Glc, Gal(beta 1-3) Gal(beta 1-3) Gal(beta 1-4) Glc, Gal(beta 1-3)[Gal(beta 1-4) GlcNAc(beta 1-6)] Gal(beta 1-4) Glc, Gal(beta 1-3) Gal(beta 1-3)[Gal(beta 1-4) GlcNAc(beta 1-6)] Gal(beta 1-4) Glc, Gal(beta 1-3)[Gal(beta 1-4) GlcNAc(beta 1-6)] Gal(beta 1-3) Gal(beta 1-4) Glc, Gal(beta 1-3)[Gal(beta 1-3) Gal(beta 1-4) GlcNAc(beta 1-6)] Gal(beta 1-4)...

Human and great ape milks contain a diverse array of milk oligosaccharides, but little is known about the milk oligosaccharides of other primates, and how they differ among taxa. Neutral and acidic oligosaccharides were isolated from the milk of three species of Old World or catarrhine monkeys (Cercopithecidae: rhesus macaque (Macaca mulatta), toque macaque (Macaca sinica) and Hamadryas baboon (Papio hamadryas)) and three of New World or platyrrhine monkeys (Cebidae: tufted capuchin (Cebus apella)and Bolivian squirrel monkey (Saimiri boliviensis); Atelidae: mantled howler (Alouatta palliata)). The milks of these species contained 6-8% total sugar, most of which was lactose: the estimated ratio of oligosaccharides to lactose in Old World monkeys (1:4 to...

Samples of milk from a Bryde's whale and a Sei whale contained 2.7 g/100 mL and 1.7 g/100 mL of hexose, respectively. Both contained lactose as the dominant saccharide along with small amounts of Neu5Ac(alpha 2-3)Gal(beta 1-4)Glc(3'-N-acetylneuraminyllactose), Neu5Ac(alpha 2-6)Gal(beta 1-4) Glc(6-N- acetylneuraminyllactose) and Neu5Ac(alpha 2-6)Gal(beta 1-4)GlcNAc(beta 1-3)Gal(beta 1-4)Glc (LST c). The dominance of lactose in the carbohydrate of these milks is similar to that of Minke whale milk and bottlenose dolphin colostrum, but the oligosaccharide patterns are different from those of these two species, illustrating the heterogeneity of milk oligosaccharides among the Cetacea. Sialyl Oligosaccharides in the Milk of Japanese Women: Changes in Concentration during the Course of Lactation OAK
The concentrations of the six major sialyl oligosaccharides found in the milk of 24 Japanese women were determined using reverse phase high performance liquid chromatography of the 3-methyl-1-phenyl-5- pyrazolone derivatives of the oligosaccharides. The milk was collected at 4, 10, 30 and 100 days postpartum. The concentrations of all six oligosaccharides decreased during the course of lactation. The concentrations of 6'-sialyllactose, sialyllacto-N-tetraose b and disialyllacto-N-tetraose decreased only after 10 days postpartum, whereas those of sialyllacto-N-tetraose a and sialyllacto-N-neotetraose c as well as 3'-sialyllactose decreased from 4 days postpartum. Large differences were observed between present data and those previously reported for Italian...

コモンウオンバットミルクオリゴ糖の構造解析 OAK
Hirayama, Kentaro; Taufik, Epi; Kikuchi, Megumi; Nakamura, Tadashi; Fukuda, Kenji; Saito, Tadao; Newgrain, Keith; Green, Brian; Messer, Michael; Urashima, Tadasu.
Previous structural characterizations of marsupial milk oligosaccharides have been performed in the tammar wallaby, red kangaroo, koala, common brushtail possum and the eastern quoll. To clarify the homology and heterogeneity of milk oligosaccharides among marsupial species, which could provide information on their evolution, the oligosaccharides of wombat milk carbohydrate were characterized in this study. Neutral and acidic oligosaccharides were isolated from the carbohydrate fractions of two samples of milk of the common wombat and characterized by H-1-nuclear magnetic resonance spectroscopy. The structures of six neutral saccharides were found to be Gal(1-4)Glc (lactose), Gal(1-3)Gal(1-4)Glc (3'-galactosyllactose), Gal(1-3)Gal(1-3)Gal(1-4)Glc...
